1. How can I contact CaptureSelect customer service?

When is the invoice send?The invoice will be send by regular mail as soon as the order is shipped, and normally arrives around the same day as the package. For all other invoice questions please contact us. 10. What are the delivery timings?The order will be processed immediately and the delivery date can be found in the order confirmation. Normally delivery takes place within 10 working days. 11. Can I cancel an order?An order can be canceled but this has to be done before the product has been shipped. To cancel send a reply to your order confirmation e-mail (automatically detailing all ordering information). Please clearly state ‘Cancellation’ and explain your reasons for cancelling the order. We may calculate an administration fee for the cancellation. The order has not been cancelled until you have received a confirmation of the cancellation.

1. What about BioProcess? The antibody toolbox affinity matrices are intended for research applications. If your are interested in using the Human Fc affinity matrix and the Fab Kappa affinity matrix for BioProcess applications please click here. What is the advantage of CaptureSelect human Fc compared to Protein A?CaptureSelect human Fc is specific for human IgG and binds all four human IgG subclasses (IgG 1-4) with high affinity, while Protein A does not bind IgG3. 3. What is the specificity of the different products from the antibody toolbox affinity matrices?The CaptureSelect antibody toolbox consists of four different affinity matrices with different specificities. CaptureSelect Human Fc: Is specific for human IgG and recognizes all four subclasses. Besides this application, the matrix is ideal for the purification of recombinant human Fc fusion proteins. CaptureSelect Multi Species IgG: Is specific for IgG and recognizes IgG of at least 17 different species. CaptureSelect Fab kappa: Is specific for human kappa light chains and binds human IgG, IgA, IgM, and IgE with kappa light chains. Also free kappa light chains are bound by this matrix. CaptureSelect Fab lambda: Is specific for human lambda light chains and binds human IgG, IgA, IgM, and IgE with lambda light chains. Also free lambda light chains are bound by this matrix. When the Fab lambda and Fab kappa affinity matrices are used in combination this will result in full human Ig purification. 4. How to perform affinity chromatography with CaptureSelect antibody toolbox matrices?1) Before use, the affinity media must be equilibrated into a suitable binding buffer. A good general binding buffer is PBS, pH 7.2-7.4 (physiological pH & ionic strength). Ultimately, the most suitable binding conditions need to be determined empirically. 2) Sample can be applied on the column. The optimal binding condition for samples is around physiological pH. Samples with acidic pH should be avoided, since binding of the target molecule will not occur at these conditions. The amount of sample that can be loaded is depending on the concentration of the target molecule in your sample and the dynamic binding capacity of the matrix. The dynamic binding capacity for human IgG is 10 mg per ml matrix. 3) After sample application, the column should be washed with binding buffer until baseline has been re-established on the monitor. A typical wash is 5-10 column volumes. 4) Elution of the target molecule from the affinity media is achieved by an acidic elution buffer, for example 0.1 M Glycine pH 3.0. If the elution is not complete or the elution peak shows tailing, elution with 0.1 M Glycine pH 2.0 may solve this problem. For CaptureSelect Fab kappa affinity matrix 0.1 M Glycine pH 2.0 is recommended. 5) The affinity matrix should be re-equilibrated in binding buffer and is ready to use for a second affinity purification run. If the column will not be used immediately, the matrix should be stored in 20% ethanol at 4 °C (39 °F). 5. How to neutralize the elution fractions of the antibody toolbox affinity matrices?After the acidic elution of the bound proteins from the antibody toolbox affinity matrices, the elution fractions can be neutralized with 1.0 M Tris pH 8.0 by adding 0.1 volume of the elution fraction. This results in 100 mM Tris pH 8.0, final concentration. The 10 times concentrated neutralizing buffer can also already be added to the tubes in which the elution fractions will be collected, which shortens the time that the eluted protein is exhibited to low pH. |
