Life Science Research products are available to solve any problem in Antibody purification by using BAC’s Antibody Toolbox ligands. These specifically designed ligands are conveniently immobilized on high quality sepharose resins. Unlike the common bacterial coat protein matrices (Protein A, G and L), the Antibody Toolbox matrices were specifically designed to be able to purify with high affinity and by using gentle elution conditions any antibody format. The Antibody Toolbox comprises seven unique products: human Fc, Fab kappa etc.

 - CaptureSelect® Human Fc   

This affinity matrix is specifically designed to purify all human IgG’s (including IgG3) and Fc fusion proteins from cell culture and transgenic expression systems, as well as from human serum and plasma.  The matrix is based on a proprietary ligand that recognizes all four subclasses of human IgG through high affinity binding at the Fc region of the human IgG heavy chain.  Unlike protein A this product has a high specificity for human IgG’s, providing the ability to separate human IgG populations (both monoclonal and polyclonal) from transgenic expression systems.  Unlike protein A this product also can purify human IgG3 (in addition to human IgG 1, 2 and 4), and can be used to purify polyclonal human IVIG from plasma and serum.

- CaptureSelect® Fab kappa

 This affinity matrix is specifically designed to purify all human kappa Ig light chain (subclass I, II, III  and IV) containing antibody formats and fragments, including kappa Fab, kappa Fab2’, and all subclasses of human kappa IgG, IgA, IgD, IgE, and IgM.  The matrix is based on a proprietary ligand that recognizes human kappa light chains through high affinity binding at the light chain constant domain.  Unlike protein L which binds the variable domain and ignores a significant fraction of the kappa light chain population, this product binds 100% of human kappa light chains.  Unlike protein L this product has a high specificity for human kappa light chains, providing the ability to separate human kappa light chain populations from transgenic expression systems.  This product can also be used to elute under gentle elution conditions.

- CaptureSelect® Fab lambda

This affinity matrix is specifically designed to purify all human lambda Ig light chain (subclass I, II, III  and IV) containing antibody formats and fragments, including lambda Fab, lambda Fab2’, and all subclasses of human lambda IgG, IgA, IgD, IgE, and IgM.  The matrix is based on a proprietary ligand that recognizes human lambda light chains through high affinity binding at the light chain constant domain. This product has a high specificity for human lambda light chains, providing the ability to separate human lambda light chain populations from transgenic expression systems. This product can also be used to elute under gentle elution conditions.

- CaptureSelect® Multi Species Fc

This affinity matrix is specifically designed to purify IgG’s from multiple species including mouse, cow, rabbit, rat, goat, horse, human and sheep.  For a complete list of species refer to the product sheet.  The matrix is based on a proprietary ligand that recognizes multiple species’ IgG’s through high affinity binding at a highly conserved epitope on the Fc region of the heavy chain.  Unlike protein G which is known to present challenges under elution conditions that result in poor yield, dissociated homodimers, and denatured antibodies, this product can be used with gentle elution conditions, retaining the structural and functional integrity of the eluate.

- CaptureSelect® Human IgA

The human IgA affinity matrix provides superior performance over other IgA purification resins in terms of specificity, purity, and dynamic binding capacity. The ligand is coupled to NHS-activated Sepharose 4 Fast Flow. This matrix can be used for the purification and isolation of human IgA from complex sources like plasma, serum, and cell culture supernatants.

- CaptureSelect® Human IgM

The CaptureSelect IgM affinity matrix contains a 14 kDa Llama antibody fragment recognizing IgM. The used ligand is directed towards a unique domain on the Fc part of IgM that enables purification of human and mouse IgM’s without cross reactivity with human and mouse IgG and IgA. This makes the CaptureSelect IgM affinity matrix a superior resin in comparison with other IgM matrices in terms of specificity and purity. The ligand is coupled to NHS-activated Sepharose 4 Fast Flow. This matrix can be used for the purification and isolation of IgM, without cross-binding to other immunoglobulins, from complex sources like plasma, serum, and cell culture supernatants.

 

- CaptureSelect® Human IgG4

This CaptureSelect affinty resin is designed to uniquely recognise human IgG4. The resin contains a 12 kDa Llama antibody fragment ligand directed towards a unique domain on the Fc part of IgG4. This enables purification of human IgG4 without cross reactivity with human IgG subclasses 1, 2 or 3. CaptureSelect IgG4 affinity matrix has a unparalleled specificity allowing one step purification. The ligand, coupled to NHS-activated Sepharose 4 Fast Flow, also shows no cross-binding with other immunoglobulins, making it suitable for IgG4 purification and isolation from complex sources like plasma, serum, and cell culture supernatants.
