Custom ligands

CaptureSelect® technology enables new opportunities in the field of custom designed ligands. BAC has developed an unique ligand discovery program that is applicable to a wide variety of target molecules, ranging from bacteria and viruses to proteins, carbohydrates and even small molecules like haptens and peptide tags.

In addition to selectivity, the robustness of the screening process also allows incorporation of key features that relate to the final purification process, for instance binding and elution conditions, contamination in the source material or ligand stability. This combination of selectivity and process functionality has proven to be highly effective in identifying ligands that meet customer’s requirements regarding defined purification processes.

Three main steps of a Phase 1 Custom Ligand Project:

Step 1 Construction of antibody expression libraries that represent the single domain antibody repertoire of a llama immunized with the target of interest.

Step 2 Screening of immune libraries, at monoclonal level, for target specific ligands and meet defined process conditions.

Step 3 Small Scale Affinity Chromatography, immobilization of purified ligands onto a suitable matrix. Followed by testing of the affinity matrix under defined process conditions for suitable chromatographic characteristics. Identified lead(s) evaluated by customer at small scale.
 
Process scale-up studies can be addressed in a subsequent Phase 2 project, which includes e.g. re-cloning of ligand lead(s) into BAC’s propriety production system in S.cerevisae.
